3. Use steps
1. Experiment preparation: Dissolve the various components in the kit on ice before use, mix gently and centrifuge briefly, then place on ice for later use;
2. Sample processing: sample processing according to "SN/T3639-2013 Campylobacter jejuni test";
3. Reagent preparation: experimental reaction system (25 μL), 21 μL qPCR mixed reaction solution, 4 μL template;
4. Reaction conditions: 95°C 3min 1cycle; 94°C 10s, 50°C 30s 40cycle;
5.Fluorescence channel: FAM;
4.Result judgment
1. The sample has a typical amplification curve, and the Ct value is less than or equal to 35;
2. The sample has no amplification curve, and the Ct value is ≥40 or not detected;
3. If the t value of the sample is >35 and <40, it needs to be re-measured. Retest results Ct value ≥ 40 is negative, Ct value < 40 is positive.
5. Storage and transport
1. The various components in the kit were dissolved on ice before use, completely mixed and centrifuged briefly, and then placed on ice for later use;
2.In order to ensure the sensitivity of the detection, it is recommended that the qPCR mixed reaction solution be frozen and thawed no more than 3 times;
3.Sample processing, reagent preparation, and sample addition should be performed in different areas to avoid cross-contamination;
