| Subculture procedure | 1. Preparation of host bacteria liquid: Take BNCC305635(ATCC11303) Escherichia coli and culture on NA plate, grow a single colony at 37 ℃ for 18-24h, pick a single colony in a liquid triangular flask (NB medium), and culture in a shaker (37 ℃ 140r /min) until OD600 is between 0.1-0.4 (2-3h) for later use; 2. Phage culture: BNCC 259107(ATCC11303-B1) Escherichia coli phage T1 freeze-dried powder was taken, 0.5mLNB was added for dissolution, all of them were injected into the host bacteria solution between 0.1 and 0.4, shaken well, let stand for 0.5h, and then placed in a shaking table (37 ℃ 170r /min) for 24h. |
