Clostridium leptum Moore et al. 1976
Culture medium Blister meat culture medium: peptone 30.0g, beef powder 3.0g, yeast powder 5.0g, soluble starch 2.0g, glucose 3.0g, sodium dihydrogen phosphate 5.0g,pH value 7.0 - 7.4,25 ℃, 1000ml distilled water is added, heated to dissolve and stirred continuously, and boiled for 1 minute. Take the minced meat residue or blistered beef grains and divide them into 15mm × 150mm test tubes about 2-3cm high, and divide the above liquid culture medium into each tube about 1cm above the surface of the meat residue or blistered beef grains. High pressure sterilization at 121 ℃ for 15 minutes for later use.
Subculture procedure (1) Prepare a test tube containing 5~10mL of liquid  medium and 2 plates; (2) open it in the biosafety cabinet, heat the tip of ampoule in the flame, quickly drop sterile water to creak it, and break it with forceps; (3) draw 0.5mL of liquid culture medium into a freeze dried ampoule, fully rehydrate and transfer the solution to the liquid test tube, mix evenly; (4) inoculate a plate with 0.2mL of suspension liquid, repeat the step to obtain two plates; ⑤ Put all the liquid test tubes and plates under the above culture conditions for cultivation, and the strains can be used when they grow.
