Subculture procedure Samples required a total of 500mL of sterile water or sterile saline to dissolve. Before use, place the quality control sample at room temperature for 30min, then open it in the biosafety cabinet, immediately add 3~4mL of sterile water, suck it out after dissolution, and put it into a sterile bottle. Repeat washing the inner wall of the vial with the remaining sterile water, and transfer it to the above sterile bottle, which is the standard quality control sample, equivalent to 500mL of water sample to be tested. The recommended filtration volume is 25~65mL, the number of fecal coliform colonies is 20~60, and the total number is less than 200.
