| Growth conditions | LB + antibiotics, 37°C |
| Subculture procedure | ① Prepare one sterile water vial and two 90mm resistant plates; ② Disinfect ampoule surface, open in biosafety cabinet, cauterize top with alcohol lamp, then quickly add sterile water to rupture; break ampoule with forceps;③ Pipette 0.5 mL sterile water into the lyophilized tube. After complete dissolution, spread the solution onto antibiotic plates at 200 μL/Agar plate; ④ Incubate antibiotic plates under specified conditions for 18–24 hours before observation (do not seal or wrap plates);⑤ Pick a single colony and inoculate it into 50 mL of LB antibiotic-supplemented liquid medium. Incubate overnight at 37°C on a shaking incubator. ⑥ Depending on experimental requirements, aspirate an appropriate amount of bacterial suspension for plasmid extraction. |
| Storage conditions | 4°C, 30 days; -20°C, 12 months. |
