Description YM agar medium
Composition Yeast Extract Powder:3.0g, Malt Extract:3.0g, Glucose:10.0g, Casein Peptone:5.0g, Agar:20.0g, pH:6.2±0.2(25℃)
Growth conditions 30°C; 24-48h; aerobic
Subculture procedure ① Prepare two plates as above; ② Disinfect ampoule surface, open in safety cabinet, cauterize top with alcohol lamp, then rapidly add sterile water to rupture. Crush with forceps; ③ Pipette 0.5mL sterile water into lyophilized tube. Thoroughly dissolve powder, then inoculate two plates (200μL/Agar plate) with uniform spread; ④ Incubate plates under specified conditions until colonies emerge.
