| Subculture procedure | ① Prepare two plates as described above; ② Disinfect the ampoule surface, open it in a safety cabinet, cauterize the top with an alcohol lamp, then quickly add sterile water to rupture it. Break it open with forceps; ③ Aspirate approximately 0.5 mL sterile water (previously equilibrated in anaerobic conditions for 24 hours) into the freeze-dried vial. Thoroughly dissolve the bacterial powder, then inoculate 2 plates with 200 μL each, spreading evenly; ④ Incubate plates under the specified conditions. |
