| Subculture procedure | ① Thaw the cryovial in a water bath (at 37°C) and shake vigorously to resuspend the bacteria; ② Disinfect the outer wall of the cryovial with an alcohol swab, then transfer it to a safety cabinet for handling; ③ Unscrew the tube cap, aspirate the entire contents, and inoculate 1–2 agar plates (approximately 200 μL per plate; for anaerobic bacteria, the medium must be placed in an anaerobic environment 24 hours in advance to remove oxygen); ④ After uniform streaking, transfer to the aforementioned culture conditions for incubation. |
