| Subculture procedure | ① Prepare 1–2 fresh agar plates as described above (pre-incubated in an anaerobic environment for 24 hours); ② Open in a safety cabinet, scorch the top with a酒精灯, then quickly add a drop of sterile water to crack the surface, and subsequently crush it with forceps; ③ Pipette 0.5 mL of liquid medium or sterile water (previously placed in an anaerobic environment for 24 hours to remove oxygen) into the freeze-dried vial, dissolve thoroughly, and mix well; ④ Pipette 0.2 mL of the bacterial suspension onto the agar plate and spread evenly; repeat twice to obtain two plates; ⑤ Incubate under the aforementioned conditions; the strain is ready for use once colonies appear. |
