Description Columbia blood agar plate
Composition Special Peptone:23.0g, Soluble Starch:1.0g, NaCl:5.0g, Agar:10.0g, Defibrinated Sheep Blood:5%(added at 50℃), pH:7.3±0.2(25℃)
Growth conditions 37°C; 24–48 h; Anaerobic
Subculture procedure ① Prepare 1–2 of the above agar plates (place in an anaerobic environment 24 hours in advance to remove oxygen); ② Open in a safety cabinet, scorch the top with a酒精 lamp, then quickly add a drop of sterile water to crack the surface, and subsequently crush it with forceps; ③ Pipette 0.5 mL of sterile water (previously placed in an anaerobic environment for 24 hours to remove oxygen) into the freeze-dried vial; after thoroughly dissolving the bacterial powder, dispense approximately 200 μL onto each of the two plates and spread evenly; ④ Incubate the plates under the aforementioned conditions; the culture is ready for use once colonies appear.
