| Culture medium | BNCC352241 |
| Description | Columbia blood agar plate |
| Composition | Special Peptone:23.0g, Soluble Starch:1.0g, NaCl:5.0g, Agar:10.0g, Defibrinated Sheep Blood:5%(added at 50℃), pH:7.3±0.2(25℃) |
| Growth conditions | 37°C; 48-72h; anaerobic |
| Subculture procedure | ① Prepare 1-2 plates as described above; ② Open plates in a biosafety cabinet after surface disinfection; ③ Pipette 1-3 ml sterile water (pre-equilibrated in anaerobic conditions for 24h) onto the Agar plate. Scrape the bacterial mat from the surface using a sterile spreader to create a suspension; ④ Aspirate the suspension with a Pasteur pipette and inoculate onto fresh plates, spreading evenly; ⑤ Incubate plates under the specified conditions. |
| Storage conditions | 2-8°C |
| Safety level | 2 |
| morphology | Colony diameter: 0.5–1 mm; circular, with neat margins; opaque, grayish-white upper surface; central elevation; smooth, glossy surface; moist texture; easily lifted; Gram-negative (red); bacillus; purity: pure |
