| Culture medium | BNCC352241 |
| Description | Columbia blood agar plate |
| Composition | Special Peptone:23.0g, Soluble Starch:1.0g, NaCl:5.0g, Agar:10.0g, Defibrinated Sheep Blood:5%(added at 50℃), pH:7.3±0.2(25℃) |
| Growth conditions | 37 ℃; 3-4 days; 5% CO2 |
| Subculture procedure | ① Prepare 1-2 fresh agar plates with typical bacterial colonies; ② Dissolve the frozen storage tube in a 37 ℃ water bath, shake quickly to dissolve, wipe the outer wall of the frozen storage tube with an alcohol swab for disinfection, and then transfer it to a safe cabinet for operation; ③ Unscrew the tube cap, aspirate all the dissolved solution, pour it into 1-2 agar plates, and apply evenly; ④ Cover the agar surface with sufficient (1-4ml) tryptone soy broth medium to maintain a liquid film throughout the entire cultivation period, and prevent the culture from growing on dry surfaces. ⑤ Place the agar plate under the above cultivation conditions for cultivation. |
| Storage conditions | -80 ℃ |
| Safety level | 2 |
| morphology | Colony diameter: 1–2 mm; round with entire margins; opaque; grayish-white on the obverse; convex center; smooth surface; moist consistency; easily lifted; Gram-negative (red); rod-shaped; pure culture. |
