Description Columbia blood agar plate
Composition Special Peptone:23.0g, Soluble Starch:1.0g, NaCl:5.0g, Agar:10.0g, Defibrinated Sheep Blood:5%(added at 50℃), pH:7.3±0.2(25℃)
Growth conditions 37°C; 48-72h; anaerobic
Subculture procedure ① Prepare 1-2 fresh plates as above (pre-deoxygenated in anaerobic environment for 24h); ② Open in a biosafety cabinet, cauterize the top with , then promptly add sterile water (pre-deoxygenated in anaerobic conditions for 24 hours) to rupture it. Subsequently break it into pieces using forceps; ③ Pipette 0.5 mL liquid medium into the lyophilized vial, dissolve completely, then return to the liquid test tube and mix thoroughly; ④ Pipette 0.2 mL bacterial suspension onto the Agar plate, spread evenly; repeat twice to obtain two plates; ⑤ Incubate under the specified conditions; the culture is ready for use once growth appears.
