| Subculture procedure | ① Prepare two of the aforementioned agar plates (pre‑equilibrated under anaerobic conditions for 24 hours).② After surface disinfection, open the ampoule in a biosafety cabinet. Flame the top of the ampoule with an alcohol burner, then quickly apply a drop of sterile water to crack it, and gently break the ampoule using sterile forceps.③ Aspirate 0.5 mL of sterile water (pre‑equilibrated under anaerobic conditions for 24 hours) and inject it into the lyophilized vial. Thoroughly resuspend the bacterial pellet, then inoculate 200 µL onto each of the two plates and spread evenly.④ Incubate the plates under the specified culture conditions. The culture is ready for use once growth is observed. |
