| Subculture procedure | ① Prepare one 18mm test tube of liquid medium, approximately 10mL (placed in anaerobic environment for 24 hours of deoxygenation in advance); ② Dissolve the Frozen vials in a water bath (fungi at 30 ℃, bacteria at 37 ℃) and shake rapidly to dissolve; ③ Wipe the outer wall of the Frozen vial with an alcohol swab for disinfection, and then transfer it to a safe cabinet for operation; ④ Unscrew the cap of the tube, aspirate all the dissolved solution, pour it into the liquid culture medium of the test tube, and place the liquid test tube under the specified culture conditions. If the bacterial solution is turbid or there is a large amount of bacterial growth at the bottom, it can be used. |
