The colonies measure 0.5-1 mm in diameter, are circular with regular margins, and are opaque. They exhibit a pale yellow pigmentation on the surface, have a raised center, and are smooth and moist in texture, making them easy to lift. The organisms are Gram-positive (G⁺, appearing blue-violet) and rod-shaped (bacilli).

Description Enhanced Clostridium agar medium
Composition Yeast Extract:3.0g, Beef Extract Powder:10.0g, Glucose:5.0g, Pancreatic Casein Peptone:10.0g, NaCl:5.0g, Soluble Starch:1.0g, L-Cysteine Hydrochloride(C3H7NO2S.HCl):0.5g, CH3COONa:3.0g, Agar:12.5g, pH:6.8±0.2(25℃)
Growth conditions 30 ℃; 24-48 hours; anaerobic
Subculture procedure ①Prepare 1-2 of the above plates (placed in an anaerobic environment for 24 hours of deoxygenation in advance) with typical bacterial colonies; ② Open the safety cabinet, burn the top with an alcohol lamp, quickly drip sterile water to break it, and then use tweezers to crush it; ③ Take 0.5mL of sterile water (placed in an anaerobic environment for 24 hours of deoxygenation in advance) and transfer it into a freeze-drying tube. After fully dissolving the bacterial powder, transfer it into a Agar plate at a rate of 200 μ L per Agar plate and apply evenly; ④ Place the petri dish under the above cultivation conditions for cultivation, and once the bacterial strain grows, it can be used.
Storage conditions 2-8 ℃
