Description Nutrient Agar
Composition Peptone:10.0g, Beef Extract Powder:3.0g, NaCl:5.0g, Agar:15.0g, pH:7.3±0.1(25℃)
Growth conditions 37°C; 18-24h; aerobic
Subculture procedure ① Prepare 1-2 plates as above; ② Disinfect ampoule surface, open in safety cabinet, cauterize top with alcohol lamp, then rapidly add sterile water to rupture. Crush contents with forceps; ③ Pipette 0.5mL sterile water into lyophilized tube. Thoroughly dissolve powder, then dispense 200μL/well onto plates, spreading evenly; ④ Incubate plates under specified conditions until colonies emerge.
