| Subculture procedure | Host culture: 37 ℃, aerobic, nutrient agar medium, 18-24h. Nutrient agar medium: 3.0g beef extract, 10.0g peptone, 5.0g NaCl, 20.0g agar (excluding liquid medium), 1.0L distilled water, pH 7.0. Sterilize at 121 ℃ for 15 minutes. Phage culture: ① Host bacterial preparation: Select a single colony of host bacteria from a Agar plate, dissolve it in 40-50mL nutrient broth medium, shake at 37 ℃, 160-180 rpm until OD600 is 0.1-0.4; ② Transfer: Take an appropriate amount of bacterial solution and transfer it into a triangular bottle of host bacterial solution; ③ Cultivation: Place the triangular flask at 37 ℃ and incubate overnight on a shaker at 160-180rpm; ④ Filtration: After 16-18 hours, centrifuge the phage culture at 4000g for 10 minutes, filter the lysate using a 0.22 μ m sterile filter, and store the filtrate at 4 ℃ |
