Description Mycoplasma Broth without Glucose
Composition Porcine gastric digest:10g, Beef extract:5g, Yeast extract:5g, NaCl:2.5g, Phenol red:0.02g, Horse serum:200ml, Penicillin:800, 000 units, pH:7.6±0.2(25℃)
Growth conditions 37 ℃, 5-7 days, aerobic culture
Subculture procedure ① Prepare 9-10 test tubes containing 4.5mL of prepared Mycoplasma culture medium (with added serum and additives); ② The Frozen vial was rapidly shaken and dissolved in a 37 ℃ water bath. The surface alcohol disinfection box was placed in a safe cabinet and opened. Then, 0.5mL of Frozen vial solution was taken into the first liquid test tube and mixed well; ④ Take 0.5mL of suspension from the first test tube and transfer it to the second test tube; ⑤ Repeat the above operation and dilute the gradient to the last tube (note: one tube needs to be left as a blank control). ⑥ Cultivate under the above culture conditions, and the strain can be used once it grows (the color of the culture tube changes from red to yellow).
